To investigate the key mechanisms behind OS cell Dox resistance, we established Dox-resistant cell lines, SaOS-2 and U2OS, by administering gradient concentrations of the drug. MTT assays revealed that the IC50 values for SaOS-2 and U2OS were 61.4 and 58.5 µM, respectively, while the resistant variants, SaOS-2-Re and U2OS-Re, displayed IC50 values of 122.9 and 118.6 µM, confirming the successful establishment of resistance (Fig. 1A).
