Editor’s summary Despite being activated by the same receptors and sharing considerable homology, SMAD2 promotes proinflammatory TH17 cell differentiation, whereas SMAD3 drives anti-inflammatory Treg cell generation. Zhang et al. uncovered a SMAD-specific posttranslational modification. SMAD2, but not SMAD3, underwent dynamic palmitoylation that promoted its phosphorylation and activation, as well as its interaction with the transcription factor STAT3, leading to TH17 cell generation in vitro.
