Abstract Cisplatin is a DNA-targeting chemotherapeutic. We have utilized a forward chemical genetics strategy to map cisplatin-damaged genes (CDGs) in A549 human lung cancer cells. Importantly, we found that cisplatin targets 77% of protein kinase genes and 96% of protein phosphatase genes in the human genome, suggesting that cisplatin can reprogram protein phosphorylation genome-wide.

Abstract Cisplatin is a DNA-targeting chemotherapeutic. We have utilized a forward chemical genetics strategy to map 7585 cisplatin-damaged genes (CDGs) with a fold-enrichment of >12 from A549 human lung cancer cells. The highly associated signalling pathways of the CDGs include sperm motility, molecular mechanism of cancer, and protein kinase A signalling. Among the CDGs, there are 1330 enzyme, 747 transcription regulators and 486 transporter genes.

Open-flow Microperfusion Combined with Mass Spectrometry for in vivo Liver Lipidomic Analysis At present conventional microdialysis (MD) technique cannot efficiently sampling lipids in vivo, possibly due to the high mass transfer resistance and/or the serious adsorption of lipids on the semi-permeable membrane of MD probe. In vivo monitoring of lipids could be of great significance for the study of disease development and mechanism.
