Alpha particles are characterized by a high linear energy transfer, which results in efficient double-strand breaks (DSBs) in DNA, and a short path length of 50–100 μm in tissues. (1) Thus, tumor-selective accumulation of alpha emitters is pivotal for achieving reasonable efficacy and safety in targeted alpha therapy. Astatine-211 (211At) is an alpha-emitting halogen applicable for cancer treatment.

1 INTRODUCTION Antibody–drug conjugates (ADCs) are an emerging class of antibody treatment for cancer. Antibody–drug conjugates consist of a tumor-targeting mAb, a highly potent payload, and a linker that connects the payload to the mAb. Specific binding of the mAb to a tumor antigen contributes to the selective delivery of the payload to cancer cells. Typically, ADCs target antigens expressed on the membranes of cancer cells.
