AAVS1‐targeted, stable expression of ChR2 in human brain organoids for consistent optogenetic control

We created human forebrain organoids that stably express ChR2 throughout entire regions of the organoids during long-term culture, which showed a non-virus mediated, robust neural activation upon light stimulation and further induced strong and consistent muscle contraction in the hybrid assembloid platform, being connected to spinal cord organoids and skeletal muscles spheroids.
