Actionable genetic landscape and dynamics in circulating DNA from 12,827 patients with metastatic breast cancer
Abstract Plasma cell-free DNA (cfDNA) is a sensitive, noninvasive tool for monitoring the genomic landscape, heterogeneity, and evolution of cancer, aggregating genetic information from multiple metastatic sites. Here, we study 15,564 cfDNA samples from 12,827 patients with metastatic breast cancer (MBC). We classify samples, deep-sequenced with Guardant360, into MBC subtypes using our trained random forest model.
Genomic biomarkers of ipatasertib response in patients with hormone receptor-positive metastatic breast cancer

Abstract AKT inhibitor biomarkers remain undefined in metastatic breast cancer (MBC). The phase Ib TAKTIC trial evaluated ipatasertib with endocrine therapy, or with fulvestrant plus palbociclib, in endocrine-resistant HR + /HER2- MBC. Exploratory baseline ± post-progression circulating-tumor DNA (ctDNA) was analyzed for genomic alterations associated with outcomes. Progression-free survival (PFS) was assessed using Kaplan-Meier and Cox modeling.

Progression-Free Survival in Metastatic Breast Cancer by Local Investigators vs Blinded Independent Central Review : A Systematic Review and Meta-Analysis
Key Points Question Is there a difference in progression-free survival (PFS) estimates when assessed by blinded independent central review (BICR) vs local investigator assessment in randomized clinical trials of hormone receptor–positive/ERBB2-negative metastatic breast cancer?
