IRG1/itaconate rewires macrophage and lung tumor metabolism through G6PD inhibition
Keywords lung cancer tumor-associated macrophages itaconate glucose-6-phosphate dehydrogenase Introduction Lung cancer is the most frequently diagnosed malignant disease worldwide and is associated with the highest mortality rate.1 In recent decades, lung cancer mortality has declined due to reduced smoking, improved detection, and the emergence of immunotherapy.
GM-CSF downregulates type I IFN responses in glioblastoma-associated monocytes
Abstract Glioblastoma (GB) is the most aggressive type of brain cancer with a devastating prognosis. Myeloid cells, especially monocytes and macrophages, comprise the majority of immune cells within the GB tumour microenvironment, where they contribute to the development of an immunosuppressive milieu hindering potential anti-tumour immune responses. Type I IFNs usually facilitate a pro-inflammatory shift in monocytes and macrophages and allow for anti-tumour functions.
Necroptosis triggers inflammatory interferon signatures in patient-derived metastatic breast cancer organoids
Abstract Breast cancer (BC) is the most common type of cancer among women worldwide and underlies relapse, disease progression, and metastasis. Resistance to chemotherapy and programmed cell death (PCD), including apoptosis, strongly affects BC therapy success and remains a major challenge.

RUNX1 interacts with lncRNA SMANTIS to regulate monocytic cell functions - Communications Biology
Abstract Monocytes, the circulating macrophage precursors, contribute to diseases like atherosclerosis and asthma. Long non-coding RNAs (lncRNAs) have been shown to modulate the phenotype and inflammatory capacity of monocytes. We previously discovered the lncRNA SMANTIS, which contributes to cellular phenotype expression by controlling BRG1 in mesenchymal cells. Here, we report that SMANTIS is particularly highly expressed in monocytes and lost during differentiation into macrophages.
Identification of a novel form of caspase-independent cell death triggered by BH3-mimetics in diffuse large B-cell lymphoma cell lines - Cell Death & Disease
Abstract BH3-mimetics represent promising anti-cancer agents in tumors that rely on the anti-apoptotic function of B-Cell Lymphoma 2 (BCL2) proteins, particularly in leukemia and lymphoma cells primed for apoptosis. Mechanistically, BH3-mimetics may displace pro-apoptotic binding partners thus inducing BAX/BAK-mediated mitochondrial permeabilization followed by cytochrome c release, activation of the caspase cascade and apoptosis.
miR-147b mediated suppression of DUSP8 promotes lung cancer progression - Oncogene
Abstract Dual-specificity phosphatase 8 (DUSP8) plays an important role as a selective c-Jun N-terminal kinase (JNK) phosphatase in mitogen-activated protein kinase (MAPK) signaling. In this study, we found that DUSP8 is silenced by miR-147b in patients with lung adenocarcinoma (LUAD), which correlates with poor overall survival.

ADPGK‐AS1 long noncoding RNA switches macrophage metabolic and phenotypic state to promote lung cancer growth
Materials and Methods Cell lines Human cell lines A549, BEAS-2B, H1650, HEK293-T, and THP1 were purchased from the American Type Culture Collection (ATCC, Manassas, USA) with cell-culture conditions per ATCC guidelines. A549 and HEK293-T cells were cultured in Dulbecco's modified Eagle's medium supplemented with 10% fetal calf serum (FCS) and 1% penicillin/streptomycin (P/S). H1650 cells were cultured in RPMI 1640 medium, supplemented with 10% FCS and 1% P/S.
Immune modulation through secretory autophagy
Recognition of EVs EVs can bind to surface receptors of recipient cells and start intracellular signaling cascades, or they can be internalized by the recipient cells, releasing their cargo.

The prostaglandin E2 receptor EP3 controls CC-chemokine ligand 2-mediated neuropathic pain induced by mechanical nerve damage
Prostaglandin (PG) E is an important lipid mediator that is involved in several pathophysiological processes contributing to fever, inflammation, and pain. Previous studies have shown that early and continuous application of nonsteroidal anti-inflammatory drugs significantly reduces pain behavior in the spared nerve injury (SNI) model for trauma-induced neuropathic pain.

GM-CSF in murine psoriasiform dermatitis: Redundant and pathogenic roles uncovered by antibody-induced neutralization and genetic deficiency
Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a pleiotropic, Th17-derived cytokine thought to critically contribute to the pathogenesis of diverse autoimmune diseases, including rheumatoid arthritis and psoriasis. Treatment with monoclonal antibodies that block GM-CSF activity is associated with favorable therapeutic effects in patients with rheumatoid arthritis.
