New N-phenylpyrrolamide inhibitors of DNA gyrase with improved antibacterial activity† This study presents the discovery of a new series of N-phenylpyrrolamide inhibitors of bacterial DNA gyrase with improved antibacterial activity. The most potent inhibitors had low nanomolar IC50 values against Escherichia coli DNA gyrase (IC50; 2–20 nM) and E. coli topoisomerase IV (22i, IC50 = 143 nM).

Introduction Structurally diverse macrocycles are of great interest due to the variety of useful properties [1]. Crown ethers have attracted particular interest over the past few decades as effective ligands for metal ion [2] and reagents for chemical analysis [3]. The ability to form complexes with various metals makes crown ethers promising extractants for extraction of various metal ions [4] including rare earths [5], rhodium [6] and other noble metals [7].
