Hope for Patients With Metastatic Bladder Cancer A new immunotherapy and chemotherapy combination has been given accelerated approval as treatment for patients with metastatic bladder cancers who cannot receive cisplatin-based treatment. The unique combination is of enfortumab vedotin (also known as Padcev) and the drug pembrolizumab (also known as Keytruda). Patients who cannot receive cisplatin for their cancer have historically fared very poorly.

Abstract Background The EV-201 trial (NCT03219333) demonstrated a clinically meaningful and durable response rate and a tolerable safety profile with enfortumab vedotin (EV) in patients with locally advanced/metastatic urothelial carcinoma (LA/mUC) treated with prior PD-1/PD-L1 inhibitor therapy and platinum-containing chemotherapy (cohort 1). Patient-reported outcome (PRO) measures were included in EV-201 as exploratory endpoints.

We acknowledge that excluding five patients from the efficacy analysis of our trial 1 Balar AV Kamat AM Kulkarni GS et al. Pembrolizumab monotherapy for the treatment of high-risk non-muscle-invasive bladder cancer unresponsive to BCG (KEYNOTE-057): an open-label, single-arm, multicentre, phase 2 study. Lancet Oncol. 2021; 22: 919-930 appears to be a departure from our protocol.
