Alphavirus infection triggers selective cytoplasmic translocation of nuclear RBPs with moonlighting antiviral roles
Keywords RNA virus protein-RNA interactions RNA-binding proteins host-virus interaction nuclear-cytoplasmic shuttling viral RNA U2 snRNP U2 snRNA spliceosome RNA in order of priorities Introduction Alphaviruses include arthropod-borne viruses (arboviruses) that can cause disease in humans, such as chikungunya virus (CHIKV), Venezuelan equine encephalitis virus (VEEV), Ross River virus (RRV), Semliki Forest virus (SFV), and Sindbis virus (SINV).1,2,3,4 These arboviruses grow in the salivary...
XRN1 supplies free nucleotides to feed alphavirus replication
Abstract Several RNA viruses induce widespread degradation of cellular mRNAs upon infection; however, the biological significance and mechanistic details of this phenomenon remain unknown. Here, we make use of a model alphavirus, Sindbis virus (SINV), to fill this knowledge gap. We found that SINV triggers cellular RNA decay through the exonuclease XRN1 and the 5-to-3 degradation machinery (5-3DM).
Absolute quantitative and base-resolution sequencing reveals comprehensive landscape of pseudouridine across the human transcriptome - Nature Methods
Abstract Pseudouridine (Ψ) is one of the most abundant modifications in cellular RNA. However, its function remains elusive, mainly due to the lack of highly sensitive and accurate detection methods. Here, we introduced 2-bromoacrylamide-assisted cyclization sequencing (BACS), which enables Ψ-to-C transitions, for quantitative profiling of Ψ at single-base resolution. BACS allowed the precise identification of Ψ positions, especially in densely modified Ψ regions and consecutive uridine sequences.
