The progression of a primary tumour to metastatic disease is a major cause of cancer-related mortality, and responses to immune checkpoint blockade (ICB) are often poor in patients with metastatic cancer. Although lymph nodes (LNs) are central hubs for immune responses, their beneficial or detrimental role in cancer progression is often disputed. Therefore, understanding how LNs influence metastatic spread is essential to improve therapy responses and patient outcomes.
