Mavoglurant reduces cocaine use in patients with cocaine use disorder in a phase 2 clinical trial
Editor’s summary Cocaine use disorder is associated with dysregulated glutamate neurotransmission. Preclinical studies have indicated that modulation of glutamate signaling by antagonizing metabotropic glutamate receptor 5 (mGluR5) can attenuate cocaine use in animal models. Gomez-Mancilla et al. conducted a randomized, placebo-controlled phase 2 clinical trial of mavoglurant, a selective noncompetitive antagonist of mGluR5, in 68 patients with chronic cocaine use disorder who also used alcohol.
