Abstract INTRODUCTION RESULTS DISCUSSION MATERIALS AND METHODS Acknowledgments Supplementary Materials REFERENCES Information & Authors Metrics & Citations View Options References Figures Tables Media Share This article has 0 eLetter . Abstract CRISPR activation (CRISPRa) enables precise up-regulation of gene expression for ex vivo and in vivo applications. However, a lack of scalable, high-coverage tools has limited comprehensive genetic screening in murine models.
A tunable Cas12a platform for single-cell perturbation screening and CRISPRi
Abstract Single-cell perturbation (Perturb-seq) screens have primarily relied on Cas9 for inducing loss-of-function phenotypes, whereas Cas12a, despite its unique effectiveness for multiplex guide expression, remains underexplored. This may be due to Cas12a’s guide RNA array (pre-crRNA) self-processing activity and the subsequent challenges associated with pre-crRNA sequence recovery during single-cell RNA sequencing library preparation.

A CRISPR/Cas9 screen reveals proteins at the endosome-Golgi interface that modulate cellular anti-sense oligonucleotide activity - Nature Communications
Abstract Anti-sense oligonucleotides (ASOs) are modified synthetic single-stranded molecules with enhanced stability, activity, and bioavailability. They associate with RNA through sequence complementarity and can reduce or alter mRNA expression upon binding of splice site positions. To target RNA in the nucleus or cytoplasm, ASOs must cross membranes, a poorly understood process.
Africa Technology Regulatory Update: Adoption of the AfCFTA Protocol on Digital Trade
The African Continental Free Trade Area (“AfCFTA”) has emerged as a pivotal opportunity that will set the framework for future trade across Africa. Amid the prospects, one of the challenges has been the fragmented and diverse regulatory environment, coupled with regulators adopting policies that are not conducive for multinationals to make investments and operate efficiently across Africa.
Systematic discovery of neoepitope–HLA pairs for neoantigens shared among patients and tumor types - Nature Biotechnology
Abstract The broad application of precision cancer immunotherapies is limited by the number of validated neoepitopes that are common among patients or tumor types. To expand the known repertoire of shared neoantigen–human leukocyte antigen (HLA) complexes, we developed a high-throughput platform that coupled an in vitro peptide–HLA binding assay with engineered cellular models expressing individual HLA alleles in combination with a concatenated transgene harboring 47 common cancer neoantigens.

What Companies Need to Know About World Bank and African Development Bank Debarment (and How to Avoid It)
In March 2017, World Bank Group president Jim Yong Kim announced that the World Bank would provide a record $57 billion in financing for projects in sub-Saharan Africa in the 2018–2020 fiscal year period. Consistent with that commitment, the World Bank’s most recent annual report indicates that $19.8 billion was issued to partner countries and businesses in sub-Saharan Africa in the 2018 fiscal year.
Non-canonical reader modules of BAZ1A promote recovery from DNA damage
Decreasing BAZ1A expression by means of RNAi sensitizes transformed human cells and primary human fibroblasts to DNA damage15,16,17, 22. In contrast, BAZ1A-deficient mouse somatic cells extracted from Baz1a −/− mice appear to respond indistinguishably from wild-type cells to DNA damage33. This prompted us to revisit the role that BAZ1A might play in responding to DNA damage in human cells by means of genetic removal. Compliance Risks from Local Content Requirements – Considerations for Doing Business in Africa
Over the last several decades, Foreign Direct Investment (FDI) by multinational companies has become a critical engine of economic growth in Africa, with FDI in the extractive industries particularly significant. A common response by local governments in Africa to increased FDI is “local content” requirements, which are designed to ensure the participation of the local population in economic activity flowing from FDI.
Ubiquilin1 is uniquely required for cell cycle entry and proliferation downstream of B-cell receptor signaling.
Ubiquilins are a family of ubiquitin-like proteins thought to be chaperones for proteasome-bound ubiquitinated substrates. Recently, ubiquilins have been implicated in neurodegenerative disease, as UBQLN2 mutations cause ALS in some humans and variants in the ubiquitously expressed UBQLN1 have been associated with Alzheimers disease. The roles for ubiquilins in immune function, however, are unknown.
