Machine-Guided Dual-Objective Protein Engineering for Deimmunization and Therapeutic Functions
Abstract Cell and gene therapies often rely on the expression of exogenous proteins derived from nonhuman organisms. An emerging consensus is to reduce the potential immunogenicity of such therapies by instead using human protein domains. However, as we engineer these human-derived proteins, we create nonhuman peptides at the linkers or junctions between domains and at mutated residues within them, which still pose a risk of immunogenicity that has largely been left unaddressed.

Assisted Fermentation by a Modified Bacillus subtilis Strain Producing Protease Improved the Quality of Sufu
1 Introduction Sufu, or furu, a traditional fermented soybean product originating from the Han Dynasty in China, is a popular flavor enhancer and appetizer because of its unique flavor, abundance of nutrients, and long shelf life (Han, Rombouts, and Nout 2001; Xu et al. 2020). Sufu can be prepared at home via the following processes.
Enantioselective photoenzymatic C(sp3)-H functionalization via single-electron oxidation of carbanions
Abstract C-H functionalization provides unparalleled benefits for the late-stage modification of complex molecules. In recent years, photocatalysis has progressed significantly due to its mild conditions, sustainability and selectivities. Current mechanisms for photocatalytic C-H functionalization primarily involve direct single-electron oxidation and direct hydrogen atom transfer (d-HAT) by radicals, both of which are invalid for the functionalization of highly electron-deficient C(sp3)-H bonds.

Multi-purpose controllable protein generation via prompted language models
Abstract Deep learning is increasingly powerful for designing proteins that meet structural and functional requirements. However, most existing methods follow a conventional pipeline: first defining a backbone structure and then generating sequences consistent with it. This approach, which encodes all design goals indirectly through structures, restricts flexibility and struggles to address multiple, complex design objectives simultaneously.

Analysis of Hydrogen Production Potential from Water Electrolysis in China
Currently, the cost of electricity for electrolysis of producing hydrogen is about 70–90% of the total cost. Scientists predict that the share of renewable energy in total energy is expected to reach about 70% in 2050, as the cost of wind photovoltaic power generation in China is as low as 0.13¥/(kW·h), and that the use of renewable energy sources, mainly photovoltaic and wind power, of producing hydrogen can effectively reduce the cost of electricity.
Three-dimensional heterogeneity in liquid electrolyte structures promotes Na ion transport and storage performance in Na-ion batteries
Three-dimensional heterogeneity in liquid electrolyte structures promotes Na ion transport and storage performance in Na-ion batteries† Mengying Ma,‡a Binbin Chen‡acd and Huilin Pan *ab Abstract Unlike solid materials, the molecular structure and chemical distribution in electrolyte solutions have been considered in isotropic states. Herein, we reveal controllable regulation of solution structures in electrolytes by manipulating solvent interactions for Na-ion batteries.

Suppressing Water Clusters by “Hydrotropic” Ionic Liquid for Highly Stable Aqueous Lithium-ion Batteries Canfu Zhang, Binbin Chen, Ziyang Cai, Fenglin Zhang, Renzhi Huang, Mengdie Yan, Yingchun Liu and Huilin Pan Abstract The state-of-the-art water-in-salt electrolytes exhibit wider electrochemical window than conventional dilute aqueous electrolytes. However, the extended electrochemical stability window via increasing salt concentration has reached a bottleneck.
Inferring gene expression from cell-free DNA fragmentation profiles - Nature Biotechnology
Abstract Profiling of circulating tumor DNA (ctDNA) in the bloodstream shows promise for noninvasive cancer detection. Chromatin fragmentation features have previously been explored to infer gene expression profiles from cell-free DNA (cfDNA), but current fragmentomic methods require high concentrations of tumor-derived DNA and provide limited resolution. Here we describe promoter fragmentation entropy as an epigenomic cfDNA feature that predicts RNA expression levels at individual genes.

Can artificial intelligence help us design vaccines?
Vaccines are among the most powerful weapons we have for preventing infectious disease. In the 1950s, hundreds of thousands of Americans were infected by measles every year. But by 2015, after decades of vaccination, a mere 191 cases were reported. Unfortunately, most vaccines take years to develop, and in the midst of a pandemic, society can’t wait. One promising approach to accelerate this process is to use machine learning, a form of artificial intelligence, to guide vaccine design.
Integrating genomic features for non-invasive early lung cancer detection
Abstract Radiologic screening of high-risk adults reduces lung-cancer-related mortality1,2; however, a small minority of eligible individuals undergo such screening in the United States3,4. The availability of blood-based tests could increase screening uptake. Here we introduce improvements to cancer personalized profiling by deep sequencing (CAPP-Seq)5, a method for the analysis of circulating tumour DNA (ctDNA), to better facilitate screening applications.
All-oxide-based synthetic antiferromagnets exhibiting layer-resolved magnetization reversal
Antiferromagnetism, a state of matter where ordered neighboring spins point in opposite directions, can be engineered in layered heterostructures, which affords control over their properties. Doing so in oxide heterostructures is tricky because the necessary ferromagnetism of the constituent layers may not survive thinning to nanometer thicknesses. Chen et al.
Natural language processing highlights treatment opportunities for rare diseases - On Medicine
Rare diseases are more common than you might think. Each individual rare disease afflicts fewer than 1 in 2000 people. But collectively, these diseases impact more than 7% of the total US population. This seeming paradox is driven by the abundance of rare diseases: more than 7,000 in total, according to the US FDA. We are not doing enough to help people suffering from rare diseases. In a recent study, we found that fewer than 4% of rare diseases had drugs approved by the US FDA in 2015.
