Neutrophil-derived apoptotic bodies function as decoy signals to preserve bone integrity during infectious osteomyelitis
Abstract Bone infection represents a pathological intersection of immunity and skeletal remodeling, where inflammatory responses drive excessive bone loss. Neutrophils dominate the early immune infiltration during infection, yet how neutrophil fate within infected bone marrow influences disease progression remains poorly understood. Here, we identify neutrophil-derived apoptotic bodies (Neut-ABs) as key mediators of bone protection during infectious osteomyelitis.
Characterizing the tumor microenvironment at the single-cell level reveals the potential immune evasion mechanism in giant cell tumor of bone
Abstract Giant cell tumor of bone (GCTB) is intermediate, locally aggressive primary bone tumor that usually occur in long bones and typically present as lytic lesions, leading to cortical expansion or destruction with a soft-tissue component. At present, the main treatment of GCTB is surgical curettage and bone cement filling, but the recurrence rate is more than 20%. It is known that tumor recurrence and immune escape are closely related to the microenvironment.
Mrgpra2 + neutrophils integrate infection-derived signals to trigger NET-mediated antimicrobial defense in bone marrow
Keywords Mrgpra2 NETs bone marrow infection signal integration Research topic(s) CP: Immunology Introduction The bone marrow is the central site of neutrophil development and functions as a reservoir for both mature and immature neutrophils, which are primed for rapid response to infectious or inflammatory stimuli.1 In response to peripheral infection, neutrophils are rapidly mobilized into the circulation and recruited to sites of tissue injury, where they exert diverse antimicrobial...
Estradiol regulates osteoclast sialylation via ST3Gal1 in postmenopausal osteoporosis - Bone Research
Abstract Estrogen deficiency after menopause accelerates bone loss by stimulating osteoclast formation and activity, but the molecular pathways that link estrogen signaling to osteoclast regulation remain incompletely defined. Here, we identify the sialyltransferase ST3GAL-I as a key mediator of RANKL-induced osteoclastogenesis. RANKL activates c-FOS to drive ST3GAL1 transcription, whereas estrogen-bound ERα competes with TRAF6 and suppresses this c-FOS–dependent induction.
Reduced osteoclast-derived apoptotic bodies in bone marrow characterizes the pathological progression of osteoporosis - Cell Death Discovery
Abstract Osteoporosis is associated with excessive activity of osteoclasts. In bone turn over, most osteoclasts undergo apoptosis after bone resorption and produce a large number of apoptotic bodies (ABs). However, the biological function of osteoclast-derived apoptotic bodies (OC-ABs) in the progression of osteoporosis is still unknow.
