To study the phenotype of mice expressing FAK Y397F or Y397E during development and in adult tissue, we targeted exon 15 of the murine FAK gene by homologous recombination in embryonic stem (ES) cells. Positive ES cell clones of each mutation were injected into blastocysts to establish the mutant knock-in (KI) mouse strains (KI neo+ ) [ 10 ].
