Highlights • single-cell POwer Simulation Tool (scPOST) is a fast and flexible simulation framework • scPOST allows users to compare study designs to assess how they affect detection power • Increasing sample size improves power more than increasing cells per sample Motivation To estimate a study design's power to detect differential abundance, we require a framework that simulates many multi-sample single-cell datasets.

New techniques analyzing single cells are being applied to clinical samples. These techniques generate large datasets, and it can be challenging to identify rare cell types associated with disease. Fonseka et al. developed a simple statistical method to do just that while simultaneously controlling for confounding biological and technical variation.
