A nested case-control study of air pollution and congenital heart defects via circulating proteins
Abstract Prenatal air pollution exposure is linked to congenital heart defects (CHD), while the underlying molecular mechanisms remain poorly understood. Here we showed that maternal exposure to PM2.5, PM10, and NO2 was significantly associated with increased CHD risk during the periconception period, with NO2 exhibiting the strongest effect. Moreover, a supervised sparse partial least square model identified pollutant-specific protein signatures in both maternal serum and umbilical cord serum.

Maternal exposure to PM2·5 and the risk of fetal congenital heart defects in China: a nationwide nested case-control study
Introduction Rapid urbanisation has intensified public health threats posed by fine particulate matter (PM2·5). In 2021, ambient PM2·5 caused 8·1 million premature deaths globally, incurring US$8·1 trillion in health costs (6·1% of the 2019 global gross domestic product).1,2 Therefore, a substantial reduction in deaths and illnesses attributable to air pollution has been listed in Target 3.9 of the Sustainable Development Goals proposed by the UN.
