Genetic landscape of stage II melanoma identifies CBL as a new driver gene and prognostic biomarker

Abstract While adjuvant therapies are currently recommended for patients with stage IIB-IV melanoma, biomarkers are missing to improve risk stratification to select the most efficient treatment and patients to be included in clinical trials. We genetically characterized a large cohort of patients with stage-II-melanoma to identify predictive and prognostic biomarkers. Clinical data of 193 stage-II-melanoma patients from the German Central Malignant Melanoma Registry were identified.

Abstract Cutaneous malignant melanoma is a common cancer in adults but extremely rare in young children, affecting fewer than one child per million each year in Europe. Because of its rarity, most treatments for children are adapted from adult therapies, despite possible biological differences. This study aimed to explore the molecular features of a rare and aggressive melanoma in a 16-month-old patient to understand disease progression and treatment resistance.

Abstract Immune checkpoint inhibitors (ICI) have significantly improved overall survival in melanoma patients. However, 60% experience severe adverse events and early response markers are lacking. Circulating tumour DNA (ctDNA) is a promising biomarker for treatment-response and recurrence detection. The prospective PET/LIT study included 104 patients with palliative combined or adjuvant ICI.
