METTL3 loss exacerbates colitis via m 6 A-dependent Slc39a8 regulation and epithelial lineage depletion
Abstract The intestinal epithelium maintains host-microbiota homeostasis, while inflammatory conditions, such as inflammatory bowel disease (IBD), induce pathological shifts in intestinal epithelial cell (IEC) subtypes. We unveil METTL3, an RNA m6A methyltransferase, as a pivotal regulator of this balance. METTL3 is enriched in intestinal stem cells and transit-amplifying cells (TACs), and upregulated in patients with IBD and a mouse model of IBD.
Evolution of limb and digit identity genes since the tetrapod ancestor
Abstract Natural selection has sculpted tetrapod limbs and digits into a tremendous morphological diversity, but the underlying developmental mechanism remains unclear. This leads to an enduring debate concerning the identity of the three digits of bird wing. Here we use comparative digit transcriptomics across six tetrapod species to examine gene expression signatures of digit evolution. Here we show that the avian wing digits are homologous to reptilian forelimb digits 1,3,4.
Convolutional neural network-long short-term memory model combined with Dammann vortex grating for decoding orbital angular momentum shift keying
This Letter presents a new artificial intelligence (AI)-based technique for decoding 16/32-ary orbital angular momentum shift keying (OAM-SK). Departing from the conventional CNN-based method that involves directly receiving vortex light at the terminal and decoding it with a convolutional neural network, our approach employs a $5\times 5$ Dammann vortex grating (DVG) to generate an OAM-SK light array (from −12th to +12th diffraction order) in the receiving plane.

MG132 inhibits proliferation and induces apoptosis of acute lymphoblastic leukemia via Akt/FOXO3a/Bim pathway
Introduction Acute lymphoblastic leukemia (ALL) is among the most prevalent pediatric cancer.1 Although recent advancements in the treatment of childhood ALL have led to significant improvements, the outcomes for patients who do not achieve long-term disease-free survival (DFS) or who experience relapse remain suboptimal.2 Given the current inadequate response to treatment in cases of refractory leukemia, it is imperative to investigate the pathogenesis of refractory and relapsed ALL and to...

Targeting m6A reader YTHDF1 augments antitumour immunity and boosts anti-PD-1 efficacy in colorectal cancer
Discussion YTHDF1 is a m6A reader is frequently upregulated in CRC, but its role in antitumour immunity is largely unknown. Here, our study demonstrates for the first time that high YTHDF1 expression in CRC drives immunosuppression. Single-cell profiling reveals that YTHDF1 is crucial to the recruitment of MDSCs, which in turn attenuated T cell infiltration and function in CRC.

Leaf extract from Vitis vinifera L. reduces high fat diet-induced obesity in mice
Leaf extract from Vitis vinifera L. reduces high fat diet-induced obesity in mice Li Meng, Yuehua Jiao, Xiaohong Zhou, Cong Liang, Kexin Yan, Ye Zhao, Xiujie Deng, Xue Han, Yanyan Yang, Hanmin Liu, Pimin Gong and Lanwei Zhang Abstract Despite the health benefits of Vitis vinifera L. leaves, its anti-obesity potential has not been fully explored. In this work, we showed that Vitis vinifera L. leaf extract (VLE) inhibits the pancreatic lipase activity.
A Phylogenetic Independent Contrast Method under the Ornstein-Uhlenbeck Model and its Applications in Correlated Evolution
New Results doi: https://doi.org/10.1101/2021.04.24.441286 Abstract Phylogenetic comparative methods are essential in studying the evolution of traits across a phylogeny. Felsenstein's phylogenetic independent contrast (PIC) method and the generalized least squares (GLS) regression were often utilized to study whether evolutionary changes between traits were correlated.
Lactiplantibacillus plantarum H-87 prevents high-fat diet-induced obesity by regulating bile acid metabolism in C57BL/6J mice
Lactiplantibacillus plantarum H-87 prevents high-fat diet-induced obesity by regulating bile acid metabolism in C57BL/6J mice Cong Liang, Xiaohong Zhou, Pimin Gong, Haiyue Niu, Linzheng Lyu, Yifan Wu, Xue Han and Lanwei Zhang Abstract Bile salt hydrolase(BSH)-producing bacteria were negatively related to the body weight gain and energy storage of the host. We aimed to obtained a novel BSH-producing strain with an excellent anti-obesity effect and explained its mechanism.
Evolution of placental invasion and cancer metastasis are causally linked
Abstract Among mammals, placental invasion is correlated with vulnerability to malignancy. Animals with more invasive placentation (for example, humans) are more vulnerable to malignancy. To explain this correlation, we propose the hypothesis of ‘Evolved Levels of Invasibility’ proposing that the evolution of invasibility of stromal tissue affects both placental and cancer invasion. We provide evidence for this using an in vitro model.
