Despite decades of study, pain is still one of the major challenges in both clinical and scientific terms. Chronic pain affects a substantial proportion of the adult population and is associated with considerable personal, social, and healthcare burdens (1, 2).

1 Introduction Advanced cancer patients often suffer excruciating pain due to bone metastases, causing bone destruction and metastatic bone pain [1]. Non-steroidal and opioid drugs commonly used in clinics only partially alleviate cancer-induced bone pain (CIBP) while exhibiting unignorable side effects [2]. Therefore, it is urgent to explore the potential mechanism of CIBP and find new therapeutic targets.

Abstract Transient receptor potential melastatin (TRPM) channels have emerged as potential therapeutic targets for cerebral ischemia–reperfusion (I/R) injury. We highlight recent findings on the involvement of TRPM channels in oxidative stress, mitochondrial dysfunction, inflammation, and calcium overload. We also discuss the challenges and future directions in targeting TRPM channels for cerebral I/R injury.
