Structural basis of fatty acid activation and transport by human FATP2
Abstract Fatty acid transport proteins (FATPs) are responsible for efficient uptake of fatty acids (FAs), essential biomolecules that play critical roles in development and growth. However, the lack of three-dimensional structures of FATPs has hindered our understanding of their mechanisms. Here, we report two cryo-EM structures of human FATP2a: wild-type (WT) FATP2a in intermediate state at 2.93 Å and a catalytic mutant of FATP2a in pre-catalytic state at 2.76 Å.

Melanoma exosomal miR-708-5p promotes macrophage M2 polarization and cancer metastasis
Abstract Monocyte-derived macrophages are usually recruited and play pivotal roles in establishing an immunosuppressive tumor microenvironment, and the interplay between tumor cells and tumor-associated macrophages (TAMs) is crucial for tumor development. However, the detailed mechanisms remain largely unelucidated in certain aggressive human cancers, such as melanoma.
CRISPR-Cas9 screening identifies ATOX1-driven cisplatin resistance mechanisms in liver cancer and evaluates targeted inhibitor efficacy - Communications Biology
|
Abstract Liver cancer treatment with cisplatin is often hindered by drug resistance. This study aimed to identify key genes associated with cisplatin resistance in liver cancer and develop targeted inhibitors. Using genome-wide CRISPR-Cas9 screening, ATOX1 was identified as a critical gene for cisplatin resistance. ATOX1 was highly expressed in liver cancer tissues and associated with poor prognosis. Knockdown of ATOX1 in liver cancer cells enhanced cisplatin sensitivity in vitro and in vivo.

Water-regulated viscosity-plasticity phase transitions in a peptide self-assembled muscle-like hydrogel - Nature Communications
Abstract The self-assembly of small molecules through non-covalent interactions is an emerging and promising strategy for building dynamic, stable, and large-scale structures. One remaining challenge is making the non-covalent interactions occur in the ideal positions to generate strength comparable to that of covalent bonds. This work shows that small molecule YAWF can self-assemble into a liquid-crystal hydrogel (LCH), the mechanical properties of which could be controlled by water.

Influence mechanism of confining pressure on the hydraulic aperture based on the fracture deformation constitutive law
Introduction Fractures are the key to fluid flow, transport, and heat transfer inside natural rock masses with distinct apertures due to the complex in situ stress (confining pressure), and knowledge of these fracture apertures plays an irreplaceable role in the seepage through fractures in many subsurface engineering applications, including seawater intrusion (Sebben et al., 2015), geothermal energy exploitation (Wang et al., 2021b; Li et al., 2022), Water–Silt Inrush Hazard (Ma et al.,...
Analysis of Surface Deformation Induced by Backfill Mining Considering the Compression Behavior of Gangue Backfill Materials
Abstract : Coal gangue, as a solid waste produced in the coal mining process, can be disposed by being prepared into backfill materials and then filled in underground goafs, thus controlling strata movement and surface subsidence. However, gangue backfill materials are non-continuous; therefore, research into the surface deformation induced by backfill mining should consider the creep compression behavior of gangue backfill materials.
