Exploring the landscape of targetable alterations in patients with glioblastoma
Abstract Glioblastomas remain the most lethal primary brain tumour in adults, with targeted therapies delivering only limited benefit despite deep molecular characterization. Several targeted drugs have received regulatory approval for low-grade gliomas, although progress in glioblastomas remains constrained by, among other aspects, extensive intratumoural heterogeneity, pathway redundancy, cellular plasticity and limited drug delivery to the central nervous system.
Single nucleus RNA profiling reveals potential therapeutic vulnerabilities in sinonasal carcinomas
Abstract Sinonasal undifferentiated carcinomas are rare, aggressive tumors with limited treatment options. Molecular subgroups defined by IDH2 mutations or SWI/SNF complex deficiencies have recently been recognized, but therapeutic implications remain unclear. We performed single-nucleus RNA sequencing on 12 FFPE tumors (six IDH2 mutated (IDH2mt), six SMARCA4 mutated (SMARCA4mt)), generating 59,612 nuclei.

Correction to: Nature Cell Biology https://doi.org/10.1038/ncb2784, published online 16 June 2013. In the version of the article initially published, the DAPI and Alexa-488 channels of the IL-8 (-4OHT) panel in Supplementary Fig. S2c and Supplementary Fig. S4a were duplicated due to an error in figure preparation. The corrected image for Supplementary Fig. S2c stemming from an existing experimental replicate is shown below as Fig. 1.

crossNN is an explainable framework for cross-platform DNA methylation-based classification of tumors - Nature Cancer
Abstract DNA methylation-based classification of (brain) tumors has emerged as a powerful and indispensable diagnostic technique. Initial implementations used methylation microarrays for data generation, while most current classifiers rely on a fixed methylation feature space. This makes them incompatible with other platforms, especially different flavors of DNA sequencing.
cIMPACT‐NOW update 10: Recommendations for defining new types for central nervous system tumor classification
Classification systems serve to group and organize data according to common relations or affinities so that they may be compared with other data. The classification system used will depend on what the classes are intended to cluster and what data are available to define these classes. Thus, the history and evolution of classifications trace the development of methods used to organize, categorize, and systematize knowledge, organisms, and objects based on shared characteristics.
Explainable artificial intelligence of DNA methylation-based brain tumor diagnostics - Nature Communications
Abstract We have recently developed a machine learning classifier that enables fast, accurate, and affordable classification of brain tumors based on genome-wide DNA methylation profiles that is widely employed in the clinic. Neuro-oncology research would benefit greatly from understanding the underlying artificial intelligence decision process, which currently remains unclear. Here, we describe an interpretable framework to explain the classifier’s decisions.
A prognostic neural epigenetic signature in high-grade glioma - Nature Medicine
|
Abstract Neural–tumor interactions drive glioma growth as evidenced in preclinical models, but clinical validation is limited. We present an epigenetically defined neural signature of glioblastoma that independently predicts patients’ survival. We use reference signatures of neural cells to deconvolve tumor DNA and classify samples into low- or high-neural tumors. High-neural glioblastomas exhibit hypomethylated CpG sites and upregulation of genes associated with synaptic integration.

AHRR and SFRP2 in primary versus recurrent high-grade serous ovarian carcinoma and their prognostic implication - British Journal of Cancer
Abstract The aim of this study was to analyse transcriptomic differences between primary and recurrent high-grade serous ovarian carcinoma (HGSOC) to identify prognostic biomarkers. We analysed 19 paired primary and recurrent HGSOC samples using targeted RNA sequencing.

Concurrent gliomas in patients with multiple sclerosis - Communications Medicine
Abstract Concurrent malignant brain tumors in patients with multiple sclerosis (MS) constitute a rare but paradigmatic phenomenon for studying neuroimmunological mechanisms from both molecular and clinical perspectives. A multicenter cohort of 26 patients diagnosed with both primary brain tumors and multiple sclerosis was studied for disease localization, tumor treatment-related MS activity, and molecular characteristics specific for diffuse glioma in MS patients.
Autumn Statement: Westfield Health Criticises Government Support for People in Work
Following the Chancellor’s Autumn Statement, Westfield Health CEO Dave Capper, says the government is failing to support employees’ health and wellbeing in the workplace. The Chancellor said many times in the lead-up to the Autumn Statement that the government was serious about tackling the long-term sickness crisis facing our country, and that helping people stay in work would be a top priority.

Multiomic neuropathology improves diagnostic accuracy in pediatric neuro-oncology - Nature Medicine
|
Abstract The large diversity of central nervous system (CNS) tumor types in children and adolescents results in disparate patient outcomes and renders accurate diagnosis challenging. In this study, we prospectively integrated DNA methylation profiling and targeted gene panel sequencing with blinded neuropathological reference diagnostics for a population-based cohort of more than 1,200 newly diagnosed pediatric patients with CNS tumors, to assess their utility in routine neuropathology.

Machine learning workflows to estimate class probabilities for precision cancer diagnostics on DNA methylation microarray data
Abstract DNA methylation data-based precision cancer diagnostics is emerging as the state of the art for molecular tumor classification. Standards for choosing statistical methods with regard to well-calibrated probability estimates for these typically highly multiclass classification tasks are still lacking.
Challenges in the histopathological classification of ganglioglioma and DNT: microscopic agreement studies and a preliminary genotype‐phenotype analysis - Blümcke - - Neuropathology and Applied Neurobiology - Wiley Online Library
Low‐grade epilepsy‐associated brain tumours (LEAT) are the second most common cause for drug‐resistant, focal epilepsy, i.e. ganglioglioma (GG) and dysembryoplastic neuroepithelial tumours (DNT). However, molecular pathogenesis, risk factors for malignant progression, and their frequent association with drug‐resistant focal seizures remain poorly understood. This contrasts recent progress in understanding the molecular‐genetic basis and targeted treatment options in diffuse gliomas.

Inadequate funding for physical and mental health
With workplace-related stress, illnesses and mental health issues becoming a bigger concern than ever, is it time for employers to be taking the burden from the NHS? More than half of working adults believe that UK businesses are not doing enough to support the physical and mental wellbeing of their employees, according to a new study released today. Contributor David Capper, Commercial Director – Westfield Health.

Working carers need more support
Supporting working carers not only boosts employee wellbeing but it can raise productivity too, says David Capper, executive director of Westfield Health The Sandwich Generation is defined as those people – typically in their thirties and forties – who care for ageing parents while also supporting their own children. This responsibility can be a cause of stress, and as a result may affect performance in the workplace.
