Are you ready? FDA has begun to implement the use of artificial intelligence in its operations, particularly during inspection activities. The FDA's new guidance on plausible mechanisms represents the first regulatory guidance-driven example of where the primary information source of a risk analysis is shifted from data-based to mechanism-based evidence.

Prognostic Significance of Incipient Ulceration in Primary Cutaneous Melanomas
Key Points Questions Should primary cutaneous melanomas with features suggestive of imminent ulceration be classified as ulcerated or as nonulcerated, per the American Joint Committee on Cancer (AJCC) definition?

Prognostic modeling of cutaneous melanoma stage I patients using cancer registry data identifies subsets with very‐low melanoma mortality
Abstract Background Evidence exists that escalating melanoma incidence is due in part to overdiagnosis, the diagnosis of lesions that will not lead to symptoms or death. The authors aimed to characterize subsets of melanoma patients with very-low risk of death that may be contributing to overdiagnosis. Methods Melanoma patients diagnosed in 2010 and 2011 with stage I lesions ≤1.0 mm thick and negative clinical lymph nodes from the Surveillance, Epidemiology, and End Results database were selected.
Characterizing the Clinical Implications of Histologic Regression in Melanoma Requires Clear Diagnostic Criteria That Are Consistently Applied
To the Editor The detailed analysis of large melanoma cohorts by El Sharouni and colleagues1 seems to settle a controversy, affirming that regression in melanoma is associated with improved prognosis. However, ambiguities in the histologic diagnosis of melanoma regression leave unanswered questions regarding its clinical implications. The lack of an assuredly consistent diagnostic approach to regression inevitably weakens the association with prognosis.

Histologic features of melanoma associated with germline mutations of CDKN2A, CDK4, and POT1 in melanoma-prone families from the United States, Italy, and Spain
Background CDKN2A, CDK4, and POT1 are well-established melanoma-susceptibility genes. Objective We evaluated melanoma histopathology for individuals with germline mutations of CDKN2A, CDK4, and POT1. Methods We assessed histopathology for melanomas diagnosed in melanoma-prone families (≥2 individuals with melanoma) from the United States, Italy, and Spain.

Acute Photodistributed Exanthematous Pustulosis Associated With Liraglutide Treatment
Acute generalized exanthematous pustulosis (AGEP) is an uncommon severe cutaneous reaction, which most commonly occurs after initiation of treatment with medications such as antibiotics, and less frequently in the setting of other triggers. Cases of acute localized exanthematous pustulosis (ALEP) have been recently reported in association with multiple medications.1,2 Here, we present a case of acute photodistributed exanthematous pustulosis (APEP) in the setting of liraglutide treatment.
