Determinants of functional burden pleiotropy and gene dosage responses across human traits
Abstract Pleiotropic and monotonic effects of gene dosage are central to understanding comorbidities in developmental pediatric and psychiatric disorders, yet the underlying biological processes are not well characterized. Here we develop a functional burden analysis to investigate the association of all protein-coding copy-number variants, genome-wide, with 43 complex traits in approximately 500,000 UK Biobank participants.

Psychotropic medications and their interactions with subcortical brain volume in bipolar disorder: An ENIGMA mega-analysis - Molecular Psychiatry
|
Abstract MRI studies in bipolar disorder (BD) have yielded inconsistent findings, partly due to the varied use of psychotropic medications. This study utilised a mega-analysis approach, accounting for concurrent medication status (syndrome-based and Neuroscience-based Nomenclature (NbN) classifications), in order to assess the association of medication status with subcortical brain volumes in BD.

Structural brain abnormalities and aggression in schizophrenia: mega-analysis of data from 2095 patients and 2861 healthy controls via the ENIGMA consortium | Molecular Psychiatry
|
Abstract It is hypothesised that structural brain abnormalities in individuals with schizophrenia are associated with aggressive behaviour, but this has not been tested directly. We pooled magnetic resonance imaging and clinical data from 2095 patients and 2861 healthy control subjects across 20 sites of the ENIGMA-Schizophrenia Working Group.

Convergence and Divergence of Rare Genetic Disorders on Brain Phenotypes
Convergence and Divergence of Rare Genetic Disorders on Brain Phenotypes: A Review | Genetics and Genomics | JAMA Psychiatry | JAMA Network [Skip to Navigation] JAMA Psychiatry. 2022;79(8):818-828. doi:10.1001/jamapsychiatry.2022.1450 Importance Rare genetic disorders modulating gene expression—as exemplified by gene dosage disorders (GDDs)—represent a collectively common set of high-risk factors for neuropsychiatric illness.
Fast and powerful genome wide association of dense genetic data with high dimensional imaging phenotypes
Several approximate or exact methods have been proposed to speed up LMM-based testing. Approximate methods assume the total polygenic random effect is same for all markers under the null hypothesis of no marker effect, hence the relevant residual genetic variance component is estimated only once using all markers. In contrast, exact methods, which is the recommended LMM practice19,20,25,35,46, estimate a residual variance component conditional on each marker’s effect.

Psychotic Experiences and Neuropsychological Functioning in a Population-based Sample
Importance Psychotic experiences in early life are associated with neuropsychological impairment and the risk for later psychiatric disorders. Psychotic experiences are also prevalent in adults, but neuropsychological investigations spanning adulthood are limited, and confounding factors have not been examined rigorously.
