The PLK4 inhibitor RP-1664 demonstrates potent efficacy in neuroblastoma preclinical models through a dual mechanism of sensitivity
Abstract It was recently shown that inhibition of polo-like kinase 4 (PLK4) induces synthetic lethality in cancers with chromosome 17q-encoded TRIM37 copy number gain due to cooperative regulation of centriole duplication and mitotic spindle nucleation. We show here that chromosome 17q/TRIM37 gain is a defining feature of high-risk neuroblastoma and renders patient-derived cell lines hypersensitive to the novel PLK4 inhibitor RP-1664.

Target antigen-displaying extracellular vesicles boost CAR T cell efficacy in cell and mouse models of neuroblastoma
|
Editor’s summary The effectiveness of CAR T cell therapy for solid tumors has been limited by poor persistence due to immunosuppressive tumor microenvironments, among other factors. Here, Giudice et al. show that neuroblastoma cells released extracellular vesicles (EVs) carrying the tumor antigens glypican-2 (GPC2) and the disialoganglioside GD2. These EVs activated CAR T cells targeting these antigens.

A humanized anaplastic lymphoma kinase (ALK)-directed antibody-drug conjugate with pyrrolobenzodiazepine payload demonstrates efficacy in ALK-expressing cancers | Nature Communications
Abstract The Anaplastic Lymphoma Kinase (ALK) gene is a receptor tyrosine kinase (RTK) with expression restricted to the developing nervous system. Most neuroblastomas express native ALK protein on the cell surface and ALK is uniformly overexpressed in fusion-positive rhabdomyosarcoma and in subsets of metastatic colorectal carcinoma, melanoma, ovarian carcinoma, and breast carcinoma.
Targeting of intracellular oncoproteins with peptide-centric CARs - Nature
Abstract The majority of oncogenic drivers are intracellular proteins, constraining their immunotherapeutic targeting to mutated peptides (neoantigens) presented by individual human leukocyte antigen (HLA) allotypes1. However, most cancers have a modest mutational burden that is insufficient for generating responses using neoantigen-based therapies2,3. Neuroblastoma is a paediatric cancer that harbours few mutations and is instead driven by epigenetically deregulated transcriptional networks4.
A Run-In With Donald Rumsfeld When I Was In College Changed The Course Of My Life
Then-Secretary of Defense Donald Rumsfeld during a press conference in Australia in November 2005. (Photo: WILLIAM WEST via Getty Images) An encounter with Donald Rumsfeld, who died earlier this week, when I was 19 drove me away from a life in government service, made me cynical about what public leaders know and care about, and led me to despise Rumsfeld for decades.
The Inheritance, Matthew Lopez's epic gay play, discussed by a multigenerational group of gay men.
We assemble a multigenerational roundtable of gay men to discuss Matthew Lopez’s epic play, currently running on Broadway, which has been hailed as a spiritual successor to Angels in America. Is it really the great gay play of the century? For questions about subscriptions or your Slate Plus feed, check our FAQ.

Milton - Not the poet-though yes,
New Age guru Louise Hay’s pseudoscience harmed the AIDS generation of gay men.
When the New Age entrepreneur Louise Hay died at 90 on August 30, the internet lit up with people praising her healing powers for often-desperate physical and psychological ailments, prescribing healthy living and major doses of self-love. But the people celebrating Hay largely ignored or brushed past the pernicious side of her prescription—the place where self-love slides into self-blame. Hay’s spiritual schema had its reasons for being, and it helped some people.
