Single-agent ibrutinib at a dose of 420 mg per day was approved in 2014 for all lines of treatment (LOTs) for chronic lymphocytic leukaemia (CLL). Overall survival (OS) improved for ibrutinib versus comparator arm in two large randomised trials, although not in the ALLIANCE trial [ 4 O’Brien S.M. Byrd J.C. Hillmen P. Coutre S. Brown J.R. Barr P.M. et al. Outcomes with ibrutinib by line of therapy and post-ibrutinib discontinuation in patients with chronic lymphocytic leukemia: phase 3 analysis.
