Discovery of 1‐(arylacetyl)‐2‐phenyl‐pyrrolidine Derivatives as Novel POLθ‐pol Inhibitors via Structure‐Based Virtual Screening and Fragment Fusion
DNA polymerase theta (POLθ) has been regarded as a promising therapeutic target for the treatment of BRCA-deficient tumors. In this study, 1-(arylacetyl)-2-phenyl-pyrrolidine derivative VS-13 was identified as a POLθ-pol inhibitor hit with an IC50 value of 1.47 μM through virtual screening. The followed similarity research obtained a more potent analogue SS-5, which was hybridized with RP-6685 to get 1-(phenylacetyl)-2-phenyl-pyrrolidine derivative F-1.

Exploring the unjamming transition of meso‐mechanical shear failure behavior in asphalt mixture
1 INTRODUCTION Asphalt pavements inevitably withstand shear loads in the actual service process, resulting in the frequent occurrence of pavement distress such as cracking, stripping, and rutting (Ling et al., 2023). This exerts a detrimental influence on the properties of the road surfaces and the safety of vehicular traffic. As it stands, the design of asphalt pavement structures is predicated on the elastic layered theory system.

Involvement of TP53 in osteosarcoma - challenges and prospects
1 Introduction Osteosarcoma is a malignant tumor arising from mesenchymal tissues and displays a bimodal age distribution: the first peak occurs between ages 10 and 14, whereas the second peak appears after age 60 (1). The pathogenesis of osteosarcoma primarily involves mutations or inactivation of tumor suppressor genes, particularly TP53, and overexpression of oncogenes such as MDM2 (mouse double minute 2 protein) (2).

Azure Virtual Networks (vNet): Overview 2025
A virtual network in Azure functions similarly to an on-premises network, allowing seamless connectivity between various resources. It enables virtual machines (VMs) to connect, form systems like FARMs for internal communication, and even integrate with on-premises systems in specific scenarios. Each Azure subscription supports multiple regions, and each region can contain one or more virtual networks. These networks remain isolated from each other, even if they reside within the same region.

Engineering a Cell-Based Orthogonal Ubiquitin Transfer Cascade for Profiling the Substrates of RBR E3 Parkin
Abstract The E3 ubiquitin (UB) ligase Parkin utilizes a Ring-Between-Ring (RBR) domain to mediate the transfer of UB to its substrates to regulate diverse cellular functions, including mitochondrial quality control, cell cycle progression, metabolism programming, and the establishment of synaptic functions. Mutations affecting the E3 ligase activity of Parkin are associated with cancer and Parkinson's disease (PD).
Analysis of Wave Load Characteristics of Hovercraft Based on Model Test
1. Introduction A fully cushioned hovercraft (hereinafter referred to as “hovercraft”) is a water surface vehicle that uses air cushions as the support to hover over water. Owing to their high speed and low resistance, hovercrafts have become an important type of vessel for ocean exploration, maritime rescue, and military operations [1]. It is an irreplaceable and important piece of equipment for water surface transportation.

Constructing fine‐grained spatiotemporal neonatal functional atlases with spectral functional network learning
1 INTRODUCTION The early stage of life is increasingly recognized to be a critical period in the establishment of the foundation for later behavioral and cognitive development (Li et al., 2019; Nelson et al., 2007). Recently, numerous imaging studies (Dubois et al., 2014; Gao et al., 2009; Knickmeyer et al., 2008; Lyall et al., 2015) have emphasized the fact that the cerebral cortex undergoes exceptionally rapid growth and significant changes during early brain development.
