Integrating bulk and single-cell RNA sequencing to identify mitochondrial quality control-related key genes in rheumatoid arthritis
Abstract Mitochondrial quality control (MQC) dysfunction contributes to rheumatoid arthritis (RA), but MQC-related genes in RA remain incompletely characterized. We integrated two bulk blood transcriptomic datasets and one peripheral blood mononuclear cell (PBMC) single-cell RNA-sequencing dataset (248 RA and 88 control bulk samples; 11,719 cells from 3 RA and 3 healthy donors).
An XGBoost-SHAP classifier to distinguish asymptomatic hyperuricemia from acute primary gouty arthritis
Abstract Asymptomatic hyperuricemia (AH) is an important precursor of acute primary gouty arthritis (AGA), but not all patients with AH will progress to acute primary gouty arthritis. Given the limitations of relying solely on uric acid levels to accurately identify AH and AGA, there is an urgent clinical need for an auxiliary identification tool that integrates routine laboratory indicators, possesses discriminative ability, and is easy to interpret.
IL27RA ubiquitination by CBL E3 ligase regulates IL27 dependent immune response against EBV- induced infectious mononucleosis
Abstract Epstein-Barr virus (EBV) is a trigger of robust immune responses and infectious mononucleosis (IM). Although EBV induces interleukin-27 (IL-27) in B cells, and IL-27 receptor alpha (IL27RA) deficiency increases EBV susceptibility, the regulatory mechanisms linking E3 ubiquitin ligases such as Casitas B-lineage lymphoma (CBL) to IL27RA during EBV infection remain unknown.
Experimental and Geant4 simulation study on 60 MeV proton energy attenuation in Ta/CNT reinforced epoxy composites
ABSTRACT Lightweight and multifunctional radiation shielding materials are required for spacecraft operating in complex space radiation environments. This study provided an experimental and numerical assessment of proton attenuation in epoxy composites reinforced with carbon nanotubes (CNTs), tantalum (Ta) nanoparticles, and hybrid Ta/CNT fillers under 60 MeV proton irradiation.

Non-pharmacological interventions have been increasingly explored as potential approaches for improving psychological symptoms and sleep quality in this population. Objective: To systematically evaluate the effects of non-pharmacological interventions on depression, anxiety, and sleep quality in patients receiving MHD.
Serum homocysteine as a novel biomarker for predicting intestinal necrosis in patients with bowel obstruction: a prospective observational study
Abstract We evaluated the diagnostic accuracy of serum homocysteine (HCY) for predicting intestinal necrosis in patients with bowel obstruction and compared its performance with conventional inflammatory biomarkers. This prospective observational study enrolled consecutive patients presenting with acute bowel obstruction between Feb 2024 and Feb 2026.
AdaBoost-based differential diagnosis of gouty arthritis and rheumatoid arthritis: model construction, temporal validation, and SHAP visualization
Abstract Background: The phenotypic overlap between gouty arthritis (GA) and Rheumatoid Arthritis (RA) poses diagnostic challenges, particularly in resource-limited settings. This study aims to develop a robust, non-invasive machine learning (ML) framework based on routine hematological parameters to facilitate precise differential diagnosis and to evaluate its robustness in a temporally independent cohort.
Natural products targeting the Nrf2 signaling pathway: potential targets and intervention strategies for the prevention and treatment of colorectal cancer
Abstract Colorectal cancer (CRC) is a highly prevalent gastrointestinal malignancy worldwide, characterised by insidious onset, rapid progression, high tendency for recurrence and metastasis, and resistance to chemotherapy. It poses significant challenges for clinical management and has become a major public health issue threatening human health. Current clinical treatments for CRC remain significantly limited.

Disease-associated genetic variants can cause missense effects in tissue-specific protein isoforms
Abstract Genetic variants can cause protein-coding mutations that result in disease. Variants are typically interpreted using the reference transcript for a gene. However, most human multi-exon genes have alternative isoforms. We show that, consistent with their reduced evolutionary constraint, coding exons in alternative isoforms harbour more population variants than exons of reference isoforms, and that these variants are more likely to cause nonsynonymous mutations.

Analysis of influencing factors for progression from type 2 to type 1 in retinopathy of prematurity
Abstract Objective: To investigate factors associated with the progression of Retinopathy of Prematurity(ROP). Methods: A retrospective analysis was performed on 148 ROP infants. All cases were classified into type 1 and type 2 ROP according to the ETROP criteria, and those progressing from type 2 to type 1 were defined as progressive type (prog-type). Intergroup differences and relevant factors for prog-type were analyzed.
Targeted AAV6 gene therapy restores corneal endothelial function in three hereditary corneal dystrophies
Results The commonly used intracameral injection (traditional anterior chamber [AC] injection) in previous mouse studies requires needle entry at the limbus and the formation of a relatively long tunnel along the corneal stroma to minimize post-injection leakage (Figures 1A and 1B ).13,15,16 The limited anterior chamber volume restricts the total injection amount, and transient intraocular pressure (IOP) elevation is inevitable during the procedure.

SpaIM: single-cell spatial transcriptomics imputation via style transfer - Nature Communications
Abstract Spatial transcriptomics (ST) technologies have transformed our understanding of cellular organization but are limited by sparse signals and restricted gene coverage. To address these challenges, we introduce SpaIM, a style transfer learning model that leverages single-cell RNA sequencing (scRNA-seq) data to predict unmeasured gene expressions in ST profiles.
The HM-TARGET personalised real-time haemodynamic targets in critical care - Nature Communications
Abstract Haemodynamic management in critical care typically relies on static, population-based targets that overlook patient-specific physiology and the evolving nature of illness. We develop and validate a framework for real-time, personalised haemodynamic management using a time-dependent Cox model that integrates static and dynamic clinical data to predict survival probabilities and derive optimal heart rate and systolic blood pressure targets over time.
“It’s so normalised. Like, yeah, I got another nude today”: image-based online sexual interactions among emerging adults
ABSTRACT As digital technologies continually change, types of negative online behaviours evolve, now covering a wide range of practices, motivations, and behaviours. Research shows that emerging adults are particularly vulnerable to image-based sexual abuse online. However, relatively little is known about emerging adults’ perceptions of and experience with image-based online sexual interactions.

Precision dosing of perampanel in pediatric patients receiving oxcarbazepine combination therapy: physiologically based pharmacokinetic modeling and simulation
ABSTRACT Background The combination of oxcarbazepine (OXC) with perampanel (PER) considerably reduces the blood concentration of PER in children, decreasing the antiepileptic effect of PER. This study aimed to predict PER exposure and provide recommendations for dose adjustment in pediatric patients.

The 11β-hydroxysteroid dehydrogenase type 1 inhibitor SPI-62 prevents morbidity in a mouse model of Cushing's syndrome
Abstract 11β-hydroxysteroid dehydrogenase type 1 (HSD-1) inhibitors represent a potential therapeutic approach for patients with Cushing's syndrome, autonomous cortisol secretion (ACS), or iatrogenic glucocorticoid (GC) excess. We assessed whether the HSD-1 inhibitor SPI-62 prevents GC-associated morbidity in a mouse Cushing's syndrome model. Corticosterone (CORT) was administered to mice for 5 weeks. Animals who received CORT were randomized between vehicle and 3 SPI-62 regimens.
