Inflammatory cytokines induce new cancer dependencies
Abstract Tumor cells respond and adapt to environmental stresses that facilitate growth in hostile environments, including cytokine-mediated inflammation elicited by antitumor immunity and enhanced by immune checkpoint blockade (ICB). However, cytokine responses also induce transcriptional and cell-state changes that may predispose tumor cells to new vulnerabilities, which remain poorly explored.
In vivo CRISPR screening in head and neck cancer reveals Uchl5 as an immunotherapy target - Nature Communications
Abstract Recurrent/metastatic head and neck squamous cell carcinoma (HNSCC) is an aggressive malignancy with a significant unmet need for enhancing immunotherapy response given current modest efficacy. Here, we perform an in vivo CRISPR screen in an HNSCC mouse model to identify immune evasion genes. We identify several regulators of immune checkpoint blockade (ICB) response, including the ubiquitin C-terminal hydrolase 5 (UCHL5).
Management of paediatric benign bone tumour
Primary tumours of bone are a diverse group of lesions that range from benign to malignant neoplasms. While primary benign bone tumours are more prevalent than malignancies in children malignant bone tumours are the seventh most common type of malignancies in children, accounting for roughly 3%–5% of cancers between the ages of 0 and 14 years.
Giant cell tumour of bone: analysis of risk factors for recurrence in 354 patients
Abstract Aims Giant cell tumour of bone (GCTB) is a locally aggressive lesion that is difficult to treat as salvaging the joint can be associated with a high rate of local recurrence (LR). We evaluated the risk factors for tumour relapse after treatment of a GCTB of the limbs. Methods A total of 354 consecutive patients with a GCTB underwent joint salvage by curettage and reconstruction with bone graft and/or cement or en bloc resection.
Neuronal CaMKK2 promotes immunosuppression and checkpoint blockade resistance in glioblastoma - Nature Communications
Mice Six- to eight-week-old C57BL/6J, LysMcre, Syn1cre, and CD45.1 mice were purchased from the Jackson Laboratory. CaMKK2−/−, Tg(Camkk2-EGFP)DF129Gsat reporter mice (CaMKK2-EGFP), and CaMKK2fl/fl mice were generously provided by Luigi Racioppi (Duke University). CaMKK2−/−, CaMKK2-EGFP and CaMKK2fl/fl mice have been previously validated22. All transgenic mouse lines were derived from or have been previously backcrossed to the C57BL/6 background.
Intermuscular extremity myxoid liposarcoma can be managed by marginal resection following neoadjuvant radiotherapy
Compared with other soft tissue sarcomas, myxoid liposarcoma (MLS) occurs in younger patients, has a propensity for intermuscular locations and is highly radiosensitive. With pre-operative radiotherapy, intermuscular MLS demonstrates substantial volume reduction and can be easily separated from surrounding tissues during resection. However, it is unclear whether marginal excision of MLS is oncologically safe.

Review: A Chilly ‘Miss Sloane’ Makes a Lecture out of Lobbying
Jessica Chastain rules the Washington-based political thriller “Miss Sloane” with such steely authority that you are hard pressed to imagine another actress playing the imperious, largely unsympathetic title character. In the opening moments, Elizabeth Sloane, a formidable Washington lobbyist, legendary for her deviousness and determination to win at all costs, impassively describes her professional modus operandi.
