Abstract Dysregulated protein synthesis has been implicated in multiple neurodevelopmental, neurodegenerative, and neuropsychiatric diseases. Protein synthesis or mRNA translation is critically regulated through phosphorylation of eukaryotic elongation factor 2 (eEF2) by its kinase eEF2K. Increased eEF2K activity leads to elevated phosphorylation and inhibition of eEF2 and inhibits the elongation phase of protein synthesis.

Introduction Alzheimer’s disease (AD) is a devastating neurodegenerative disease. However, its pathogenesis and an effective treatment have yet to be further elucidated. In addition to extracellular deposition of amyloid-β peptides (Aβ) and intracellular aggregation of hyperphosphorylated tau, a number of studies have revealed that neuroinflammation is a critical event in the pathogenesis of AD (McGeer et al., 2016). Microglia and astrocytes are major cells involved in neuroinflammation.
