SIRT2-mediated deacetylation of ANT3 at Lys96 promotes cell death
Abstract ANT3 is a key molecule that constitutes the mitochondrial permeability transition pore (mPTP) and mediates cell death. Disruption or conformational changes in ANT3 protein homeostasis can trigger cell death. However, the molecular mechanism of ANT3 protein homeostasis regulation remains unclear.
Phosphorylation of OTUB1 promotes autophagy initiation under starvation
Abstract Eukaryotes initiate autophagy in response to environmental challenges such as nutrient deprivation. However, the mechanisms governing autophagy initiation remain incompletely understood. Here, we demonstrated that OTUB1 phosphorylation plays a key role in starvation-induced autophagy initiation. Specifically, ERK phosphorylates OTUB1 at S118 during starvation.
Local delivery of IL-15 and anti-PD-L1 nanobody by in vitro-transcribed circILNb elicits superior antitumor immunity in cold tumors
Keywords circCV-B3 circILNb biotin-avidin purification system IL-15 anti-PD-L1 nanobody tumor immunotherapy Introduction Immunotherapies have revolutionized the clinical paradigm of tumor treatment. Immune checkpoint inhibitors (ICIs) that target PD-1/PD-L1 continue to be approved by the Food and Drug Administration (FDA) for monotherapy and combination therapy of tumor.1,2 Nevertheless, the clinical benefits of these approaches in patients with advanced tumor have been limited.
Deacetylation of ANXA2 by SIRT2 desensitizes hepatocellular carcinoma cells to donafenib via promoting protective autophagy - Cell Death & Differentiation
Abstract Hepatocellular carcinoma (HCC) is one of the most lethal forms of cancer globally. HCC cells frequently undergo macroautophagy, also known as autophagy, which can lead to tumor progression and chemotherapy resistance. Annexin A2 (ANXA2) has been identified as a potential therapeutic target in HCC and is involved in the regulation of autophagic process. Here, we for the first time showed that ANXA2 deacetylation plays a crucial role in donafenib-induced autophagy.
F-box protein FBXO32 ubiquitinates and stabilizes D-type cyclins to drive cancer progression - Nature Communications
Abstract D-type cyclins (hereafter, cyclin D) are central regulators orchestrating G1/S cell cycle transition. Accordingly, aberrant expression of cyclin D is strongly correlated with proliferation-related diseases such as cancer. However, the mechanisms regulating cyclin D turnover are incompletely elucidated. Here we identify FBXO32, namely atrogin-1, as the E3 ubiquitin ligase that targets all three cyclin D for ubiquitination and stabilization.

Electrochemical Quinuclidine-mediated Minisci-type Acylation of N-Heterocycles with Aldehydes
Electrochemical Quinuclidine-mediated Minisci-type Acylation of N-Heterocycles with Aldehydes The electro-generation of acyl radicals from both aromatic and aliphatic aldehydes remains an unmet challenge. We provide a solution to this challenge by merging electro-oxidation and quinuclidine-mediated hydrogen atom transfer strategy. The generation of acyl radicals at decreased applied potentials than that of formyl oxidation exhibits excellent functional group compatibility.
