Discussion In this study, we identify a pivotal role for DXM in modulating anti-myeloma immunity within the bone marrow microenvironment. Under basal conditions, GR acts as a scaffold that assembles an inhibitory complex with PPP1CB, TBK1, and IRF3, keeping TBK1-IRF3 dephosphorylated and the pathway silenced. Upon DXM binding, GR undergoes a conformational change, leading to complex dissociation and release of the brake.
