Genome-directed discovery of an octalin-containing polyether ionophore improves diabetic skin wound healing
Abstract Chronic hard-to-heal skin wounds in diabetes mellitus pose a significant clinical challenge, underscoring the urgent need for additional therapeutics. Herein, we report the identification of a polyether biosynthetic gene cluster (pdm BGC) from the marine-derived Streptomyces marincola SCSIO 03032 through genome mining.
Occupational disparities in chronic disease prevalence and metabolic covariation: a cross-sectional study of six occupational groups in Lanzhou
Abstract Objectives: To investigate the differences in chronic disease prevalence and underlying metabolic profiles among working-age individuals across six typical occupational groups, identify occupation-related associated factors, and provide epidemiological evidence for occupation-specific precise prevention and control of chronic diseases. Methods: A cross-sectional study design was adopted.
ICIsAtlas reveals a suppressive NK cell niche in pan-cancer immunotherapy profiles
Abstract Immune checkpoint inhibitors (ICIs) have reshaped the treatment in multiple tumors. However, a substantial proportion of patients exhibit limited responses. The lack of harmonized, large-scale pan-cancer ICI datasets coupled with accessible analysis tools hinders the systematic discovery of response biomarkers and resistance mechanisms.
Deep generative optimization of mRNA codon sequences for enhanced mRNA translation and therapeutic efficacy - Nature Communications
Abstract Messenger RNA (mRNA) therapeutics show immense promise, but their efficacy is limited by suboptimal protein expression. Here, we present RiboDecode, a deep learning framework that generates mRNA codon sequences for enhanced mRNA translation. RiboDecode introduces several advances, including direct learning from large-scale ribosome profiling data and generative exploration of a large sequence space.
Defining ortholog-specific UHRF1 inhibition by STELLA for cancer therapy - Nature Communications
Abstract UHRF1 maintains DNA methylation by recruiting DNA methyltransferases to chromatin. In mouse, these dynamics are potently antagonized by a natural UHRF1 inhibitory protein STELLA, while the comparable effects of its human ortholog are insufficiently characterized, especially in cancer cells.
A single-dose circular RNA vaccine prevents Zika virus infection without enhancing dengue severity in mice - Nature Communications
Abstract Antibody-dependent enhancement (ADE) is a potential concern for the development of Zika virus (ZIKV) vaccines. Cross-reactive but poorly neutralizing antibodies, usually targeting viral pre-membrane or envelope (E) proteins, can potentially enhance dengue virus (DENV) infection. Although E domain III (EDIII) contains ZIKV-specific epitopes, its immunogenicity is poor.
Recognition of extremophilic archaeal viruses by eukaryotic cells: a promising nanoplatform from the third domain of life
Morphological characteristics of SMV1 and SSV2 were studied by TEM and representative micrographs are presented in Fig. 1. SMV1 is spindle-shaped with an extending tail at one pole. SMV1 appears both individually and in a rosette-like arrangement, where virus-virus interaction appears to be mediated through the tail regions. Although, the general morphology of SMV1 remains unaltered after PKH67 labelling, some virions appear more electron dense.
