Nano-enabled spatially selective protein degradation modulates lactate metabolism to potentiate antitumor immunity in liver cancer
Abstract Dysregulated cancer metabolism, driven in part by excessive lactate export through monocarboxylate transporters 1 (MCT1) and 4 (MCT4), generates an acidic tumour microenvironment that suppresses antitumour immunity and diminishes therapeutic efficacy. Although small-molecule inhibitors targeting MCT1 and/or MCT4 have been explored, their clinical translation is limited by systemic toxicity.
Speed politics and socialist contradictions in Maoist documentary cinema
Abstract During the Great Leap Forward (1958–1960), Chinese cinema found itself trapped between heightened political pressures and a fervent pursuit of shortened production cycles, using exaggerated numerical information within its cinematographic language to represent extreme labor intensity and the dreamlike speed of historical development.
High-throughput Raman-activated cell sorting of microalgal genome-wide edited library revealed a regulatory pathway for carotenoid synthesis
Abstract Functional genomics have been hampered by the paucity of efficient methods that connect genotype and metabolic phenotype at single-cell resolution. Using the industrial microalga Nannochloropsis oceanica as a model, we introduced a platform that comprises a genome-wide single-gene-edited mutant library and high-throughput Raman-activated cell sorting (RACS).
HuR coordinates systemic aging through platelet infiltration
Abstract Aging involves morphological and functional changes across different organs, but how these changes are linked among the different organs remains to be elucidated. Here, we uncover a central role of platelets in systemic aging. In aged mice, the levels of platelet-secreted pro-inflammatory factors (PSPF) increased greatly in the serum and platelets, leading to a diffuse increase of platelet infiltration in the brain, liver, lung, kidney, and aortic root.
Glucocorticoids elevate clear cell renal cell carcinoma sensitivity to HIF-2α inhibitors by suppressing H4K12 lactylation
|
Abstract Approximately 70% of clear cell renal cell carcinoma (ccRCC) patients harbor von Hippel‒Lindau (VHL) deficiency, which drives pseudohypoxia and metabolic reprogramming. Here, we report a histone H4 lysine 12 lactylation (H4K12la)-fueled phosphoglycerate kinase 1 (PGK1)-lactate positive feedback loop that sustains glycolytic flux in VHL-deficient ccRCC and is pharmacologically disruptable by glucocorticoids.

The inner nuclear membrane protein LEMD3 organizes the 3D chromatin architecture to maintain vascular smooth muscle cell identity - Nature Communications
Abstract Maintaining the contractile phenotype of vascular smooth muscle cells (VSMCs) is critical for vascular homeostasis. However, the role of the 3D chromatin architecture in regulating VSMC identity remains elusive. A genome-scale CRISPR screen identifies LEMD3 as a potential regulator to maintain VSMC identity. Lemd3 deficiency in VSMCs results in the loss of the contractile phenotype and exacerbates intimal hyperplasia in mice.

Shallow-water mussels (Mytilus galloprovincialis) adapt to deep-sea environment through transcriptomic and metagenomic insights - Communications Biology
Abstract Recent studies have unveiled the deep sea as a rich biosphere, populated by species descended from shallow-water ancestors post-mass extinctions. Research on genomic evolution and microbial symbiosis has shed light on how these species thrive in extreme deep-sea conditions. However, early adaptation stages, particularly the roles of conserved genes and symbiotic microbes, remain inadequately understood.
Pd-Catalyzed Asymmetric Etherification of 2H-chromenes: Enantioselective Construction of Chiral 4-Alkoxy-4H-chromenes
Pd-Catalyzed Asymmetric Etherification of 2H-chromenes: Enantioselective Construction of Chiral 4-Alkoxy-4H-chromenes A new strategy for the construction of chiral 4-alkoxy-4H-chromene skeleton was reported. A series of chiral 4-alkoxy-4H-chromenes containing trifluoromethyl group were obtained with good yields (up to 80% yield) and excellent enantioselectivity (up to 93% ee). Stabilization Strategies of Lithium Metal Anode Toward Dendrite‐Free Lithium‐Sulfur Batteries
Lithium-sulfur (Li-S) batteries are considered as a most promising rechargeable lithium metal batteries because of their high energy density and low cost. However, the Li-S batteries mainly suffer the capacity decay issue caused by the shutting effect of lithium polysulfides and the safety issues arising from the Li dendrites formation. This review outlines the current issues of Li-S batteries.
Inactivating histone deacetylase HDA promotes longevity by mobilizing trehalose metabolism
Abstract Histone acetylations are important epigenetic markers for transcriptional activation in response to metabolic changes and various stresses. Using the high-throughput SEquencing-Based Yeast replicative Lifespan screen method and the yeast knockout collection, we demonstrate that the HDA complex, a class-II histone deacetylase (HDAC), regulates aging through its target of acetylated H3K18 at storage carbohydrate genes.
Cellular response to moderate chromatin architectural defects promotes longevity
Abstract Changes in chromatin organization occur during aging. Overexpression of histones partially alleviates these changes and promotes longevity. We report that deletion of the histone H3-H4 minor locus HHT1-HHF1 extended the replicative life span of Saccharomyces cerevisiae. This longevity effect was mediated through TOR signaling inhibition.
