Abstract Enhancer of zeste homolog 2 (EZH2), a histone methyltransferase, has gained attention as a promising therapeutic target in osteoarthritis (OA) due to its central role in modulating inflammation, catabolism, and hypertrophy within chondrocytes. Previous studies have further demonstrated that EZH2 inhibition can slow OA progression in surgically induced mouse models, highlighting its potential in reducing joint degradation.

Discussion Understanding the pathogenesis of RA to propose new therapeutic options remains a current need more than 20 years after the avenement of TNF inhibitors.

Methods Discovery cohort: ESPOIR cohort We used the baseline data from patients with early RA included in the ESPOIR (Etude et Suivi des POlyarthrites Indifférenciées Récentes) cohort, details of which have been previously described.15 Briefly, these patients have had at least two swollen joints persisting for more than 6 weeks but less than 6 months.
