An overnight stimulation protocol with short Brefeldin A incubation is optimal to assess polyfunctional T cell responses in whole blood In order to establish an optimal ex-vivo stimulation protocol to study antigen-specific T cell responses suitable for field study sites, three different protocols (SET1, SET2 and SET3, Table 1) were compared using heparinized whole blood from healthy donors (n = 5) stimulated with staphylococcal enterotoxin B (SEB).
