Anagho et al. profiled DNA-damage-stimulated ADP-ribosylation in wild-type and BRCA-mutant breast cancer cell lines using mass spectrometry. They showed that the predominantly serine-targeted ADPr response to H2O2-induced DNA damage was homogeneous across cell lines, regardless of BRCA1/2 expression or PARP inhibitor sensitivity.

Abstract ADP-ribosylation (ADPr) signaling plays a crucial role in the DNA damage response. Inhibitors against the main PARP1 enzyme catalyzing ADPr after DNA damage, are used as targeted therapies against breast cancers with BRCA1/2 mutations. However, development of resistance to PARP inhibitors (PARPi) is a major obstacle in treating patients.
