Iberdomide Triplet Improves MRD-Negative Complete Response in Relapsed/Refractory Multiple Myeloma

Iberdomide combined with daratumumab and dexamethasone nearly doubled the rate of minimal residual disease (MRD)-negative complete response compared with daratumumab, bortezomib, and dexamethasone in patients with relapsed or refractory multiple myeloma. Lonial et al reported the primary MRD analysis of the phase III EXCALIBER-RRMM trial in The Lancet Oncology. The trial has two primary endpoints: MRD-negative complete response and progression-free survival.

Chronic Immune-Related Adverse Events Common After Adjuvant Anti–PD-1 Therapy for Melanoma

Nearly half of patients with resected high-risk melanoma treated with adjuvant anti–PD-1 therapy developed chronic immune-related adverse events, and more than one-third still had ongoing toxicity after a median follow-up of more than 5 years. Roma A. Kankaria, MD, and colleagues reported the findings in JAMA Network Open. Immune checkpoint inhibitors can produce durable benefit in melanoma, but they can also cause immune-related adverse events (irAEs) that persist after treatment ends.

Anitocabtagene Autoleucel Shows High Response Rate in Heavily Pretreated Relapsed/Refractory Multiple Myeloma

Anitocabtagene autoleucel (anito-cel), an investigational B-cell maturation antigen (BCMA)-directed chimeric antigen receptor (CAR) T-cell therapy, produced responses in all treated patients in a phase I study of heavily pretreated relapsed or refractory multiple myeloma, with 79% achieving a complete response. Frigault et al reported the findings in The New England Journal of Medicine. Anito-cel uses a synthetic d-domain binder designed to detach from BCMA relatively quickly after binding.
