Introduction Glutamate is a major type of excitatory neurotransmitters in the central nervous system and plays an important role in learning, memory, and cognition (Danbolt, 2001). Group II metabotropic glutamate receptors (Group II mGluRs) are the peri-synaptic receptor of glutamatergic neurons and mainly distribute in the presynaptic membrane (Swanson et al., 2005; Niswender and Conn, 2010).

Introduction Vascular endothelial cells in the brain are closely linked to each other through various junction proteins, and interact with pericytes and astrocytes to form a special blood-brain barrier (BBB) barrier system. BBB strictly limits the entry of neurotoxic substances, inflammatory factors and immune cells in the blood into central nervous system (CNS), and excludes the metabolites and neurotoxic substances in CNS from the brain.

Highlights Aquaporins are attractive targets for therapeutic intervention in the diverse conditions associated with water and solute dyshomeostasis that affect millions of patients worldwide every year. Aquaporin drug discovery has made little progress, possibly due to a range of assumptions including the intrinsic non-druggability of the aquaporin pore, compounded by issues with the reproducibility of current assays.
