Divergent Role for STAT5 in the Adaptive Responses of Natural Killer Cells
Highlights • STAT5 is induced in NK cells by IL-12 via STAT4 during MCMV infection • NK cells require STAT5 for protection against viral infection • IL-2 and IL-15 non-redundantly drive clonal proliferation of anti-viral NK cells • IL-15 and STAT5 promote NK cell survival during the memory phase Summary Natural killer (NK) cells are innate lymphocytes with the capacity to elicit adaptive features, including clonal expansion and immunological memory.
Transcription Factor IRF8 Orchestrates the Adaptive Natural Killer Cell Response
Natural killer (NK) cells are innate lymphocytes that display features of adaptive immunity during viral infection. Biallelic mutations in IRF8 have been reported to cause familial NK cell deficiency and susceptibility to severe viral infection in humans; however, the precise role of this transcription factor in regulating NK cell function remains unknown. Here, we show that cell-intrinsic IRF8 was required for NK-cell-mediated protection against mouse cytomegalovirus infection.

Atg5 Is Essential for the Development and Survival of Innate Lymphocytes
Autophagy Is Required for Lymphocyte Survival following Homeostatic Proliferation but Is Dispensable in Lymphoreplete Mice Induction of Autophagy by Metformin Enhances Lymphocyte Survival following Homeostatic Proliferation To study the functional relevance of increased autophagic activity in lymphocytes during lymphopenia without the confounding developmental defects, we utilized mice with either inducible deletion of the essential autophagosome machinery component Atg5 (UbcCre-ERT2 ×...
Adipose-resident group 1 innate lymphoid cells promote obesity-associated insulin resistance
Innate lymphoid cells (ILCs) are the most recently identified arm of the innate immune system that function to protect epithelial barriers against pathogens and maintain tissue homeostasis. While ILCs can also promote pathology at these mucosal sites, it remains unknown whether aberrant activation of tissue-resident ILCs can contribute to disease in non-barrier tissues.
The transcription factor IRF8 regulates natural killer cell development and response to cytomegalovirus infection
Interferon regulatory factor 8 (IRF8) is a transcription factor critical for specifying monocyte and dendritic cell lineage fate and for directing B cell development, yet its function in natural killer (NK) cells is not understood. NK cells are innate lymphocytes that develop in the bone marrow and are capable of antigen-specific clonal proliferation.
