ZNRF3 and RNF43 are active monomeric E3 ubiquitin ligases that self-associate
Editor’s summary Abstract Supplementary Materials REFERENCES AND NOTES Information & Authors Metrics & Citations Check Access References Figures Tables Media Share Editor’s summary The Wnt signaling pathway becomes aberrantly activated in many cancer types. Its activity is opposed by ZNRF3 and RNF43, RING domain–containing E3 ubiquitin ligases that target Wnt receptors for degradation. Padala et al. investigated the structural and functional characteristics of the RING domains of ZNRF3 and RNF43.
Allosteric ligands with distinct properties uncover tissue-specific physiological regulation mediated by free fatty acid receptor 2

Phosphorylation bar-coding of free fatty acid receptor 2 is generated in a tissue-specific manner
Request a detailed protocol Flp-In T-REx 293 cells harbouring hFFAR2-DREADD-eYFP were cultured to confluence. Receptor expression was induced with 100 ng/ml of doxycycline for 24 hr, followed by 5 min stimulation at 37°C with sorbic acid or MOMBA (100 μM). The cells were washed with ice-cold PBS, and TE buffer containing protease and phosphatase inhibitor cocktails was added to the dishes followed by transfer of cells into pre-chilled tubes.
