To the Editor, Secondary acute myeloid leukemia (AML), that is AML arising after prior exposure to cytotoxic therapy (tAML, therapy-related AML) and/or antecedent hematological disorder (AHD), has generally been associated with a worse prognosis in comparison to de novo AML. However, it is increasingly recognized that genetics rather than clinical history may explain these outcome differences [1-3].

Our methods and results made extensive use of data from previous landmark studies26,31. Care was taken to follow good practices in the analyses of methylome and transcriptome data, employing widely approved procedures previously used in other high-standard studies. Regarding the handling of large cohorts, we used sample correlations, performed genotype checks between omics data, and added technical and biologic replicates wherever possible.
