Hypoxic regulation of chromatin and gene transcription
Abstract Adaptation to reduced levels of oxygen (hypoxia) is an essential feature of eukaryotic life. Within the animal kingdom, cellular responses are orchestrated by the transcription factor HIF (Hypoxia Inducible Factor) which is regulated by specific 2-oxoglutarate-dependent oxygenases. This family of enzymes also includes histone demethylases, and histone methylation has also been observed to increase in hypoxia.

RAM Is Required for RNMT Interaction with RNA To define the role of RAM in transcription, RNA Pol II occupancy on the genome was investigated using chromatin immunoprecipitation (ChIP). Following transfection with RAM siRNA or non-targeting control for 36 hr, RNA Pol II ChIP was performed on three biological replicates that exhibited a high correlation genome wide and at an individual gene level (Figures S4x Download (9.98 MB ) Document S1. Figures S1–S9 and Table S1Figures S4A and S4B).
