Results To assess the consequences of Sgsh deficiency on brain ultrastructure, we analyzed frontal cortex of wild-type (WT) and Sgsh−/− mice by transmission electron microscopy (EM). In Sgsh−/− mice, we observed markedly enlarged (>1000 nm), electron-lucent vacuoles most prominently in cells bearing small, electron-dense nuclei; an ultrastructural pattern consistent with microglia and lysosomal expansion (Figures 1A IV-V, yellow arrows, and S1A).
