Abstract Global epigenetic resetting in the gonadal primordial germ cells (PGCs) enables transition from early PGCs to gametogenesis and eventual restoring of totipotency after fertilization. This reprogramming process involves global DNA demethylation, changes in nuclear morphology and remodeling of repressive histone modifications.

Introduction In sexually reproducing organisms, germ cells play a crucial role in transmitting the genetic and epigenetic information necessary for the development of a new organism, serving as an enduring link between generations (Leitch et al., 2013a; Ramakrishna et al., 2021). Primordial germ cells (PGCs), the earliest embryonic precursors of the gametes (Saitou and Hayashi, 2021), are likely specified in humans around embryonic day 12–16 (post-conceptional week 2) (Alberio et al., 2021).
