Introduction In sexually reproducing organisms, germ cells play a crucial role in transmitting the genetic and epigenetic information necessary for the development of a new organism, serving as an enduring link between generations (Leitch et al., 2013a; Ramakrishna et al., 2021). Primordial germ cells (PGCs), the earliest embryonic precursors of the gametes (Saitou and Hayashi, 2021), are likely specified in humans around embryonic day 12–16 (post-conceptional week 2) (Alberio et al., 2021).

Abstract DNA methylation is a critical epigenetic mark in mammalian cells. Many aspects of DNA methylation maintenance have been characterized; however, the exact kinetics of post-replicative methylation maintenance remain a subject of debate. Here we develop isolation of DNA by 5-ethynyl-deoxyuridine labelling for mass spectrometry (iDEMS), a highly sensitive, quantitative mass spectrometry-based method for measuring DNA modifications on metabolically labelled DNA.
