XPO1-mediated TRIM21 nuclear export reprograms TREM2+ macrophage polarization by targeting IRF3 to augment anti-PD-1 efficacy in small cell lung cancer
Abstract Small cell lung cancer (SCLC) is an aggressive malignancy that responds poorly to immune checkpoint inhibitor (ICI) therapy, due to its immunosuppressive tumor microenvironment. Here, we analyse preclinical mouse tumor models and patient-derived SCLC samples and identify Exportin 1 (XPO1) as a regulator of immune suppression in SCLC.
Nuclear to Cytoplasmic Transport Is a Druggable Dependency in HDAC7‐driven Small Cell Lung Cancer (Adv. Sci. 14/2025)
Graphical Abstract New Subtype of Small Cell Lung Cancer In article number 2413445, Dingzhi Huang, Mengjie Li, and co-workers report that a HDAC7-high small cell lung cancer (SCLC) subtype has a poor prognosis and have identified XPO1 as a druggable target in this subtype. Elevated HDAC7 predicts increased sensitivity to selinexor both in vitro and in organoids, suggesting selinexor as a promising therapeutic option for HDAC7-high SCLC patients.
Nuclear to Cytoplasmic Transport Is a Druggable Dependency in HDAC7‐driven Small Cell Lung Cancer
1 Introduction Small cell lung cancer (SCLC) accounts for ≈15% of lung cancers, and is highly aggressive with limited treatment options and poor prognosis, with a 5-year survival rate <6%.[1] The incorporation of immunotherapy into first-line platinum-etoposide chemotherapy has only modestly enhanced outcomes for SCLC patients in unselected populations.[2] Regrettably, targeted therapies have shown limited to no success in SCLC, failing to achieve the desired results, including those...
Predictive and prognostic markers from endoscopic ultrasound with biopsies during definitive chemoradiation therapy in esophageal squamous cell carcinoma | BMC Cancer | Full Text
Patients and treatments Between January 1, 2015, and December 31, 2019, a cohort of 821 patients who underwent dCRT and had not received treatment previously for EC (stage I-IVA) at the Tianjin Medical University Cancer Institute and Hospital were enrolled. Only biopsy-confirmed squamous cell carcinoma with pre- and interim-treatment EUS measurements was eligible for inclusion (Supplementary Figure S1).
