Surface expression of antitoxin on engineered bacteria neutralizes genotoxic colibactin in the gut - Nature Microbiology
Abstract Colibactin, a metabolite produced by gut bacteria carrying the polyketide synthase (pks) island, is associated with host genotoxicity and tumorigenesis. However, no Food and Drug Administration-approved therapeutics directly target colibactin. Here we show that expression of the intracellular colibactin self-resistance protein (ClbS) on the surface of engineered bacteria shields the host from genotoxic effects across multiple pks+ isolates.
Selective HLA knockdown and PD-L1 expression prevent allogeneic CAR-NK cell rejection and enhance safety and anti-tumor responses in xenograft mice - Nature Communications
|
Abstract Allogeneic cellular immunotherapy exhibits promising efficacy for cancer treatment, but donor cell rejection remains a major barrier. Here, we systematically evaluate human leukocyte antigens (HLA) and immune checkpoints PD-L1, HLA-E, and CD47 in the rejection of allogeneic NK cells and identify CD8+ T cells as the dominant cell type mediating allorejection.

Engineering innate immune cells for cancer immunotherapy - Nature Biotechnology
Abstract Innate immune cells, including natural killer cells, macrophages and γδ T cells, are gaining prominence as promising candidates for cancer immunotherapy. Unlike conventional T cells, these cells possess attributes such as inherent antitumor activity, rapid immune responses, favorable safety profiles and the ability to target diverse malignancies without requiring prior antigen sensitization. In this Review, we examine the engineering strategies used to enhance their anticancer potential.
Single-cell RNA sequencing defines distinct disease subtypes and reveals hypo-responsiveness to interferon in asymptomatic Waldenstrom’s Macroglobulinemia | Nature Communications
Abstract Waldenstrom’s Macroglobulinemia (WM) is an IgM-secreting bone marrow (BM) lymphoma that is preceded by an asymptomatic state (AWM). To dissect tumor-intrinsic and immune mechanisms of progression, we perform single-cell RNA-sequencing on 294,206 BM tumor and immune cells from 30 patients with AWM/WM, 26 patients with Smoldering Myeloma, and 23 healthy donors.
IL-12/15/18-induced cell death and mitochondrial dynamics of human NK cells
Introduction Mitochondria are dynamic organelles that coordinate several cellular functions, including metabolism, differentiation, cell death and autophagy, among others (1). The relevance of mitochondria for natural killer (NK) cell effector functions has been evidenced, especially in the context of cancer. Zheng et al.
Autologous cellular therapy for myeloma: Giving ex vivo expanded NK cells their due
Ex vivo feeder-free autologous NK cell expansion is an efficient modality for clinical translation. In this issue of Cell Reports Medicine, Nahi et al. describe the use of this approach as consolidation therapy for multiple myeloma, 1 Nahi H. Chrobok M. Meinke S. Gran C. Marquardt N. Afram G. Sutlu T. Gilljam M. Stellan B. Wagner A.K. et al. Autologous NK Cells as Consolidation Therapy Following Stem Cell Transplantation in Multiple Myeloma. Cell Reports Medicine.
Enabling clinical grade manufacturing of gene-engineered NK cells
Published: 5 January 2022 Innovator Insight In the adaptive cell and gene therapy field, T cells are currently receiving a lot of focus and showing success in making it to the market – but natural killer (NK) cells are beginning to close this gap. In particular, CAR NK cells for cancer cell therapy offer a number of distinct advantages. But what benefits, and what challenges, do CAR NK cells pose?
Enabling clinical grade manufacturing of gene-engineered NK cells
Clinical evidence has shown that natural killer (NK) cells hold the promise for successful adoptive immunotherapy for cancer. Regulated by their germ-line encoded activating and inhibitory receptors, NK cells can recognize and eliminate tumor cells rapidly without prior sensitization. To further enhance their anti-tumor efficacy, NK cells can also be modified by chimeric antigen receptors (CARs).

Human cytokine-induced memory-like NK cells expand in patients with AML and display enhanced anti-leukemia responses.
NK cells are innate lymphoid cells that mediate anti-tumor responses. Relapsed/refractory AML patients have a poor prognosis and new therapy approaches are needed. Recently, memory-like properties of NK cells have been discovered, including cytokine-induced memory-like NK cells. Human NK cells briefly pre-activated with IL-12, IL-15, and IL-18 and then allowed to differentiate in vitro have greater re-stimulation responses compared to control cells.
