Synthesis of Compounds tert-Butyl (2-(2-(4-((6-Amino-2-butoxy-8-hydroxy-9H-purin-9-yl)methyl)benzamido)ethoxy)ethyl)carbamate (1) tert-Butyl (2-(2-aminoethoxy)ethyl)carbamate (380 mg, 1.86 mmol) and DIPEA (0.523 mL, 2.99 mmol) were dissolved in DCM (0.5 mL) and added to the stirring solution of compound T7 (200 mg, 0.56 mmol) in DMSO (3 mL). To an ice-chilled solution was added COMU (0.640 g, 1.49 mmol), and the mixture was stirred at room temperature for 3 h.

Abstract Pattern recognition receptor (PRR) ligands hold great promise as adjuvants and immunotherapeutics. Here, we demonstrate that chemical conjugation of PRR ligands results in synergistic immune response amplification inaccessible to unlinked agonist mixtures.
